Efecto de la inhibición de la actividad aminopeptidasa del plasma seminal sobre los parámetros de calidad seminal en conejo

    • GnRH analogues can be absorbed via mucosa less efficiently than via parenteral. This difference may be due to the presence of enzymes in the seminal plasma. These enzymes among which aminopeptidases are included, can condition the hormone bioavailability when the induction of ovulation takes places using extenders with GnRH analogue present in them. In this context, the aim of this study was to evaluate the inhibition of aminopeptidase activity in the rabbit seminal plasma with various inhibitors (bestatin, EDTA and a protease inhibitor cocktail of Sigma-Aldrich®) and study its effect on semen quality. Results showed that the presence of inhibitors did not affect the motility neither the acrosome integrity of the samples. On the other hand, the aminopeptidase activity was inhibited by all extender tested. Extenders with bestatin and Sigma’s cocktail inhibited significantly the seminal plasma aminopeptidase activity in a greater extent than EDTA extenders (65 and 63% versus 38 and 33% for bestatin, cocktail, 20mM and 30mM EDTA, respectively). In conclusion, since apparently they do not affect sperm quality and are able to inhibited around the 60% of the seminal plasma aminopeptidase activity, the next step would be to add bestatin or the protease inhibitor cocktail to the seminal extender with the hormone and perform artificial insemination.
