Caracterización morfológica y molecular de las poblaciones de "Cryphonectria parasitica" en castaños de Galicia

      El cancro del castaño, causado por Cryphonectria parasitica, es una enfermedad ampliamente extendida en las zonas de castaño de todo el mundo. Las únicas perspectivas actuales para su control se basan en el conocimiento de las poblaciones del hongo en los sotos afectados y la aplicación de cepas hipovirulentas. Se realizó un estudio de la biología de las poblaciones de C. parasitica en castaños de las provincias de Lugo y Ourense para conocer la diversidad de los tipos de compatibilidad vegetativa (vc) y sexual, y la incidencia de cepas hipovirulentas. Se obtuvieron 610 aislados, que en cultivo, mostraron en su mayoría un fenotipo virulento. Solo cuatro aislados de la provincia de Lugo presentaron ARNbc, demostrando la presencia de hipovirulencia. La caracterización molecular de los hipovirus determinó que pertenecían al subtipo E de Cryphonectria hypovirus 1 (CHV1-E).

      La diversidad de la compatibilidad vegetativa fue muy baja. Se encontraron en total 9 tipos vc, compartiendo 5 de ellos las dos provincias. Cuatro tipos fueron compatibles con tipos vc europeos conocidos. Los otros 5 son nuevos tipos europeos que denominamos E1, E2, E5, E6 y E7. Los tipos vc dominantes en ambas poblaciones fueron E1, E2 y E5, abarcando entre los tres el 97% de los aislados. Se encontraron cuatro cepas hipovirulentas: dos compatibles con el tipo vc E1, una con E2 y otra con E5. El estudio de compatibilidad sexual mostró que el idiomorfo MAT-1 dominaba ambas poblaciones.

      La reproducción principalmente asexual, junto con la baja diversidad de tipos de compatibilidad vegetativa y la presencia de cepas hipovirulentas compatibles con los tipos vc dominantes, son factores que favorecen la aplicación de técnicas de control biológico del cancro en los castaños afectados de Lugo y Ourense.

      Chestnut blight caused by Cryphonectria parasitica is a fungal disease widely spread in chestnut stands all over the world. At present, the only feasible perspectives for its control are based on the knowledge of the populations present in the affected stands and the use of hypovirulent strains. The biology of C. parasitica populations naturally occurring in chestnut stands in Lugo and Ourense provinces was then studied so as to determine the diversity of vegetative compatibility (vc) and mating types, as well as the occurrence of hypovirulent strains. A total of 610 isolates were obtained. Only 4 isolates, from Lugo province, showed the presence of ARNbc, characteristic of hypovirulent strains. Molecular characterization of hypovirus proved all hypovirulent strains belonged to Cryphonectria hypovirus 1 (CHV1-E).

      The diversity of vegetative compatibility was very low. Nine vc types were found, five of them occurring in both provinces. Four types were compatible with the known European testers and the others were unreported vc types named E1, E2, E5, E6 and E7.

      Dominant vc types in both populations were E1, E2 and E5, being detected in 97% of the isolates. Four hypovirulent strains were found: two compatible with vc type E1, one with E2 and other with E5. The study of mating types showed that idiomorph MAT-1 was dominant in both populations.

      Reproduction mainly asexual, together with the low diversity of vegetative compatibility types and the presence of hypovirulent strains compatible with dominant vc types, are factors favouring the application of a biological control program against chestnut blight in chestnut stands in Lugo and Ourense provinces.
