Mitochondria are pivotal organelles responsible for generating ATP through oxidative phosphorylation and for regulating essential cellular processes, including apoptosis and metabolism. They possess their own genome, maintained and replicated by a specialized set of proteins known collectively as the mitochondrial DNA (mtDNA) replisome. Dysfunctions in any component of this replisome can lead to the accumulation of mtDNA mutations, depletion, or instability, ultimately compromising mitochondrial function and contributing to various mitochondrial disorders, premature aging, and neurodegenerative diseases. To investigate these mechanisms, we employed an integrative approach combining cryo-electron microscopy (cryo-EM), biophysics, and biochemistry. Furthermore, we developed novel methodologies to enhance sample preparation, providing improvements to the cryo-EM field. This thesis advances the structural and functional characterisation of the mtDNA replisome and introduces innovative strategies for cryo- EM sample preparation.
