Resumen de Integración de factores transcripcionales MYB de un dominio en rutas de transducción de señales

    El maíz es uno de los cereales más consumidos del mundo gracias a su semilla, gran fuente de hidratos de carbono, proteínas y ácidos grasos. Para la acumulación eficaz de estos nutrientes se requiere un tipo celular especial, las células de transferencia, situadas en la región basal del grano de maíz. Estas células están encargadas de aumentar la superficie de intercambio entre la parte materna y filial gracias a grandes repliegues de su pared y membrana plasmática. Conocer las señales que afectan a la diferenciación, desarrollo y mantenimiento de estas células puede derivar en importantes mejoras en el rendimiento de las cosechas.

    ZmMRP-1 es el primer factor transcripcional clonado específico de este tipo celular. Se ha demostrado que su expresión promueve la diferenciación de las células de transferencia y que en plantas transgénicas de especies heterólogas como son cebada, tabaco y Arabidopsis, el patrón de expresión de su promotor se restringe a regiones de alto intercambio de nutrientes, como en la región equivalente a la capa de células de transferencia de la base de la semilla de estas especies y en zonas de infección parasitaria. A pesar de esto, es poco lo que conocemos sobre la regulación de este gen, ya que no se han encontrado mutantes de pérdida de función y no se han podido obtener líneas transgénicas sobreexpresando la proteína ya que, aparentemente, la elevada expresión es tóxica para la planta.

    Esta tesis tiene como objetivo principal el estudio de los factores que puedan estar modulando la actividad de ZmMRP-1 mediante la obtención de plantas transgénicas de Arabidopsis capaces de sobreexpresar de manera inducible la proteína, partiendo de la hipótesis de que, dada su función y localización, es plausible su activación mediante hormonas y/o azúcares. Además pretendemos determinar si hay un gen ortólogo a ZmMRP-1 en Arabidopsis, ya que de esta manera puede que sea posible aprender sobre su regulación y función de manera más sencilla que en maíz.

    Gracias al diseño de líneas transgénicas sobreexpresoras y minigen de expresión inducible, hemos encontrado que, efectivamente, ZmMRP-1 tiene relación con hormonas, especialmente auxinas, etileno y citoquininas, si bien no hemos podido determinar si los azúcares tienen algún papel en la modulación de la actividad del factor transcripcional. Además hemos obtenido una serie de candidatos potenciales de genes ortólogos en Arabidopsis, especialmente el gen At5g04760, que presenta un patrón de expresión similar al observado para el promotor de ZmMRP-1 al ser introducido en Arabidopsis

    Corn is one of the most consumed cereals in the world, as its seed is a major source of carbohydrates, proteins and fats. In order for these nutrients to accumulate efficiently in the endosperm, it is necessary the presence of a special cell type known as transfer cells. These cells are located in the basal area of the maize kernel or BETL region. Consequently, the study of the signals that alter the differentiation, development and maintenance of these cells may derive in substantial improvements in yield.

    ZmMRP-1 was the first transcription factor to be cloned in this region, and it is specific of transfer cells. Its expression is required for the differentiation of these cells. When introduced in heterologous species like wheat, tobacco and Arabidopsis, the expression pattern is less restrained than in maize, although the promoter is functional in areas with a high rate of nutrient flow, such as those equivalent to the BETL region in the seeds of these species or in areas exposed to parasite attack. Nonetheless, little is known about the regulatory pathways that govern its expression as we have not found any knock-out mutants on the maize databases. It has also been impossible to use the knock-down or overexpression approaches as the late showed to be lethal for the transgenic plants.

    The main aim of this thesis is the study of the possible factors modulating the activity of ZmMRP1, based on the hypothesis that, given its location, it is feasible that its activation is driven by hormone and/or sugar signaling. We also tried to find the Arabidopsis ortholog gene. Its identification would facilitate the study of its function and regulation, as compared to the maize system.

    We obtained several overexpression and minigene transgenic lines in Arabidopsis, in which the activity of ZmMRP-1 is repressed by an inducible system. These lines have helped us to find that indeed, ZmMRP-1 is regulated by hormones, mainly auxins, ethylene and cytokinins, but we have not been able to demonstrate any relationship between this transcription factor and sugar signaling. We also found several potential Arabidopsis genes which might be orthologs of ZmMRP-1. It is particularly interesting the gene At5g04760, because it showed a similar expression pattern than the promoter of ZmMRP-1 when introduced in Arabidopsis. This gene will help us understand the role of this transcription factor in the species of interest, maize.
