Characterization of a partial sequence of the P-subunit of the glycine decarboxylase gene from four coffea genotypes

    • A 427 bp fragment of DNA isolated from leaf tissue of four coffee genotypes (Coffeá arabica cv. Caturra, Coffea canephora, Coffea COngenesis and Coffea eugenioides) was amplified by PCR. Using heterologous primers synthesized following the nucleotide sequence encoding a part of P-subunit of glycine decarboxylasa from pea Pisum sativum. Amplified fragments were cloned and their nucleotide sequence was determined. Sequence analysis showed 79% homology between the gene sequence of the P-subunit of glycine decarboxylase from P. sativum and the four Coffea genotypes. The amino acid sequence of cloned fragments showed 81% homology between P. salivum and the four Coffea genotypes, whereas between Flaveria pringleí and Coffea such homology was 85%. Comparison of nucleotide sequences for the. P-subunit gene from the four Coffea genotypes showed 98-100% and 99-100% homology at amino acid level, except for a substitution of glutamine by glutamic acid (position 59) in C. canephora and C. congensis.
