  • The aim was to evaluate the effect of activin-A added to the IVM and IVC media on cleavage rate, embryo development and blastocysts quality of oocytes from prepubertal goat. COCs were randomly allocated into four groups: (1) A0A0: COCs matured in IVM medium and cultured in SOF medium; (2) A0A10: COCs matured in IVM medium and cultured in SOF medium+10ng/mL activin-A; (3) A10A0: COCs matured in IVM medium+10ng/mL activin-A and cultured in SOF medium; and (4) A10A10: COCs matured in IVM medium + 10ng/mL activin-A and cultured in SOF medium+10ng/mL activin-A. Cleavage rate was recorded at 48hpi and blastocysts yield at Days 7, 8 and 9 pi. In order to assess the quality of blastocysts obtained from the treatment groups, cell number and the cellular composition of Day 8 pi blastocysts were evaluated by differential staining of ICM and TE. The cleavage rate did not show differences between treatments, however, blastocysts yield improved with the addition of activin-A. The highest blastocyst rate (18.7%) was obtained when activin-A was added only in IVC medium (A0A10). No differences in blastocyst mean cell number were detected among groups. In conclusion, the presence of activin-A during IVC improves developmental competence of prepubertal goat oocytes.
