Progesterone is essential for the preparation of the endometrium and the maintenance of pregnancy, so supplementation with progesterone after artificial insemination (AI) could increase fertility in lactating dairy cows. The aim of this study was to determine the effects of treatment with a progesterone intravaginal device (PRID) between Days 16 and 19 post-AI on the expression of the genes ISG15, Mx1 and Mx2. On Day 16 post-AI, lactating dairy cows were randomly allocated to one of two groups, control (n=5) or treatment (n=9) with a PRID for 3 days. Blood samples were collected on Day 20 after AI and the expression of genes in peripheral mononuclear cells was determined by RT-qPCR. The overall pregnancy was 50% (7/14). Cows diagnosed as pregnant had a higher gene expression of ISG15, Mx1 and Mx2 compared to those non-pregnant (p<0.05). Supplementation with progesterone post-IA did not affect the expression of ISG, Mx1 and Mx2 on Day 20 post-IA. Further studies are needed to clarify the effect of progesterone treatment on ISGs
