El análisis del efecto citotóxico se lleva a cabo con la linea celular diploide, derivada de cultivo primario de pulmón humano (Fp/6) y con la línea celular esta blecida de ovario de hámster chino (CHO). La inhibición del crecimiento se cuantí fica por contenido proteico y nivel de ATP.
Como sistema de activación metabólica se ha utilizado fracción postmitocondrial de hígado de rata (S9) previamente inducidas con Aroclor 1254.
