Resumen de Surface sterilisation of phylloxera eggs for investigating grapevine‐phylloxera interactions in tissue culture

Alison V. Kellow, Angela Corrie, Robyn van Heeswijck

  • A simple method has been developed for sterilising eggs of grape phylloxera (Daktulosphaira vitifoliae Fitch) to use in controlled experiments involving in vitro cocultivation of phylloxera with grapevine (Vitis spp.). Eggs are collected and washed with 70% ethanol followed by a sterile water rinse using a Millipore filtration apparatus. Such a filter provides a rapid means of transferring a known number of eggs into tissue culture, and subsequently determining percentage hatched.
