Los métodos para aislar e identificar mutantes defectivos en la respiración anaeróbica del nitrato (Nit-) tienden a ser más específicos para ciertos genes mutados. En el presente trabajo se desarrolló un método denominado TNC, basado en una modificación del publicado por Fimmel y Haddock (1979), pero con mayores ventajas: no fue necesario incubar en anaerobiosis estricta, permitió ampliar el espectro de genes mutados (incluyendo mol y fnr ) y detectar la aparición de colonias rojas muy rápidamente, en sólo 24 horas de crecimiento. La frecuencia de mutantes Nit- y la distribución de genes mutados fueron superiores, al usarlo comparativament e con el método de selección por resistencia al clorato.
El TNC resultó muy sensible y con reproducibilidad para id entificar diferencialmente clones mutantes Nit- (TNC+) y silvestres Nit+ (TNC-); además fue eficiente para caract erizar cepas bacterianas y en los análisis genéticos. En el presente trabajo también se estableció y evaluó un criterio de clasificación fisiológica para agrupar preliminarmente a los mutantes pleiotrópicos Nit- Gas- en cuatro subclases genotípicas, al observar que las mutaciones en moe revertían fenotípicamente con molibdato, en forma tardía.
The methods to isolate and to identify defective mutants in the nitrate anaerobic respiration (Nit-) tend to be preferential for certain mutated genes. This work presents a method denominated TNC, based in a modification of Fimmel and Haddock published in1979, but with highest advantages: it was not necessary to incubate with strict anaerobiosis, permitted both to amplify the spectre of mutated genes (including mol y fnr ) and to detect very quickly the appearing red colonies, in only 24 hours of growth. Using it comparatively with the method of selection for chlorate resistance, the frequency of Nit- mutants and the distribution of genes mutated were higher. The TNC showed to be very sensitive and with reproducibility to identify differently Nit- mutants (TNC+) and Nit+ wild type (TNC-) clones; furthermore, it was efficient for the characterisation of bacteria strains and the genetic analysis. In the present work it was also established and evaluated a criteria of physiologic classification for preliminary grouping pleiotropic Nit- Gas- mutants in four genotypic subclass, by observing that moe mutations could be reverted phenotypically in late form with molybdate.
