Vitis amurensis Rupr., distributed in the Northeast of China, is the most cold-hardy grape species. It can tolerate temperatures as low as -40~-50°C when fully cold acclimated. Our study showed that as the temperature drops, the cold hardy ?Zuoshan-1? grapevine accumulates ABA quicker and in higher concentration than less cold-hardy Vitis species. To better understand the molecular mechanisms of ABA and its role in grape cold hardiness, we constructed a suppression subtractive hybridization (SSH) cDNA library to isolate ABA inducible genes of ?Zuoshan-1?. In a total of 214 randomly picked clones from the ABA-induced library, 167 were successfully sequenced, from which 31 contigs were assembled. Blast analysis revealed that 14 contigs were homologous to Vitis or other plant species while the other 17 contigs showed high homology to Escherichia coli, which apparently resulted from E. coli contamination. Functional analysis indicated that the putative functions of these contigs were associated with photosynthesis, cell defense or detoxification, and signal transduction.
