Grapevine genotypes with combined downy and powdery mildew resistance

    • The resistant cultivar ?Panonia? and breeding lines from the species V. rotundifolia were cross bred and backcrossed to V. vinifera L. with the aim of creating new grapevine cultivars with good fruit quality and higher resistance to downy and powdery mildew. This way, resistance specificities from different genetic sources would be combined into cultivars with more robust resistance against a wider range of pathogen strains. A few seedlings with the highest level of downy and powdery mildew resistance were phenotypically selected after several years of field observations in unsprayed vineyards.

      The aim of this research was to determine the presence of known downy and powdery mildew resistance genes in the selected genotypes. Based on the pedigree, we assessed the inheritance of Rpv3, Rpv12, Run1/Rpv1, Ren3 which were present in the ancestors of these selections. Genomic DNA was extracted using a CTAB-based method from a young leaf. For PCR reaction we used published primers that generate polymorphic markers tightly linked to the genes.

      Two breeding lines tested positive for the presence of resistant alleles at all tested genes. One of them bears female flowers, it cannot be recommended for cultivation but it represents a convenient maternal parent for breeding - not requiring emasculation prior to blooming. The other is under evaluation for agronomic characteristics and wine quality. We provide further evidence of the strong predictive power of marker-assisted selection for Rpv3, Rpv12, Run1/Rpv1, Ren3.
