Resumen de Caracterización metabólica y enzimática de la resistencia al herbicida nicosulfuron en biotipos de rottboellia cochinchinensis (lour.) w.D. Clayton

    Rottboellia cochinchinensis es una maleza de gran importancia y su control químico se basa principalmente en el uso del herbicida nicosulfuron; sin embargo, se ha observado una disminución del efecto del herbicida en el control de la maleza, la cual ha mostrado algunos índices de resistencia. Con la finalidad de caracterizar metabólica y enzimáticamente la resistencia de poblaciones de R. cochinchinensis al herbicida se hicieron una serie de experimentos que consistieron en la confirmación de la resistencia del biotipo seleccionado, a través de un análisis de regresión log-logística que permitió determinar el índice de resistencia (IR). El estudio de la naturaleza de la resistencia se realizó a través de una prueba de inhibición del complejo P 450 monooxigenasas con el uso de malatión, y una prueba enzimática in vivo donde se midió por colorimetría la acumulación de acetoína, como una medida indirecta de la inhibición de la acetolactato sintetasa (ALS). El bioensayo de respuesta a dosis permitió confirmar la resistencia del biotipo (IR = 3,87). No se observó una reducción de la biomasa aérea fresca de las plantas resistentes tratadas con malatión con respecto a las no tratadas, indicando que las isoenzimas citocromo P 450 monooxígenasas que podrían ser inhibidas por el malatión no participaron en la degradación del nicosulfuron en este bioensayo. En la prueba enzimática in vivo se demostró la insensibilidad de la enzima ALS del biotipo resistente al herbicida nicosulfuron, lo que permitió concluir que el mecanismo de resistencia de este biotipo es por el sitio de acción del herbicida.

    Rottboellia cochinchinensis is a major weed and its chemical control is mainly based on the use of the herbicide nicosulfuron;

    however, there has been a decrease in the effect of the herbicide on the weed which has showed some levels of resistance. In order to perform a metabolic and enzymatic characterization of the resistance of R. cochinchinensis to nicosulfuron, some experiments were conducted to confirm the selected biotype resistance through an analysis of log-logistic regression and determine the resistance index (IR). The study of the resistance nature was done through inhibition tests of the P 450 monooxygenases complex by the use of malathion, and in vivo enzymatic assay where acetoin accumulation was measured by colorimetry as an indirect measure of the inhibition of ALS. The dose-response bioassay confirmed the resistance (IR = 3.87). There was no reduction in fresh biomass of resistant plants treated with malathion compared to the untreated ones, indicating that cytochrome P 450 monooxygenase isozymes that could be inhibited by malathion were not involved in the degradation of nicosulfuron in this bioassay. With the in vivo enzymatic assay, we demonstrated the insensitivity of the ALS enzyme of the resistant biotype to nicosulfuron, therefore it is concluded that the resistance mechanism of this biotype is in the action site of the herbicide.
