This experiment introduces the Poisson distribution to undergraduate analytical chemistry students through the use of single-molecule spectroscopy/microscopy. Students obtain fluorescence microscopy images of single dye molecules dispersed in an optically transparent polymer film. The spatial distribution of molecules in the image is studied through a grid analysis, and the results are modeled with the Poisson distribution. The spatial concentration of the sample is also considered. Potential adaptations of the experiment to more common imaging instrumentation and non-single-molecule samples are presented.
