Review of contraindicated drug-drug interactions, excluding antiretrovirals

    • The interactions between drugs are possible whenever a person takes two or more medications concurrently. There are a number of mechanisms by which drugs interact with each other and can be divided into two main categories: pharmacokinetic and pharmacodynamic interactions.

      The aim of this study was to conduct a literature review of clinically relevant drug interactions, excluding antiretrovirals, to build a list of clinically relevant drug-drug interactions. The list will contain the medicines that interact classified as «contraindicated», the mechanism of the interaction and the effect involved.

      A systematic literature search was conducted on PubMed using free text terms,reference lists from studies and reviews identified were searched manually for relevant studies.Based on the literature obtained above, a list of clinically relevant interactions was built. The list was then double-checked with other databases such as Micromex® and the on-line application CheckTheMeds®.

      In this paper we developed a list of 164 drug interactions that were categorized as major drug-drug interactions and contraindicated drug-drug interactions frequently found in hospitals, the antiretrovirals were excluded due to their high number of interactions found and they will be included in another paper.

      Due to the high number of localized interactions, we also, restricted the table to only consider the use of contraindicated interactions to prevent the phenomenon called «alert fatigue». Finally a table with 34 drug interactions classified as contraindicated,excluding antiretrovirals, for use in hospitals and electronic prescribing programs was developed
