The objective of the present research was to purify the early pregnancy factor (EPF) from swine serum using diafiltration and ion-exchange chromatography and compared with that obtained from seudopregnant serum of PAF intrauterine administrated sows. EPF activity was determined by using rosette inhibiton titre (RIT) after the pregnancy serum obtained from a mature swine 13 days after mating, or pseudopregnancy serum from a PAF intrauterine administered sow. When the pattern of SDS-page of pregnant serum was compared with the bands obtained from non-pregnant swine serum (day 0 of cycle, estrus) the 30 KDa band was not confirmed in the non-pregnancy serum. This band (30 KDa) also appeared in pseudopregnancy serum from PAF intrauterine administered sow.When this fraction (30 KDa) was applied onto the DEAE-Sepharose column, the EPF activity was only found in the unabsorbed fraction. Next this fraction was applied onto the S-CL6B sepharose column. No EPF activity was found in the different eluates. It is concluded that EPF is a protein with 30KDa of molecular weight and it has a cationic structure.
