Respuesta humoral murina y humana contra antígenos solubles de huevo y excreción-secreción de gusanos de Schistosoma mansoni, tratados con praziquantel

    La respuesta inmune y el efecto del praziquantel (PZQ) sobre los adultos S. mansoni son sinérgicos. Se evaluó la IgG e IgE en suero de ratones infectados con cercarias provenientes de infección unimiracidial en caracoles B. glabrata (infección UNI) y en pacientes infectados, contra antígenos soluble de huevo y productos de excreción-secreción de gusanos hembras y machos incubados o no con PZQ in vitro, mediante western blot. La IgE en el suero de ratones con infección UNI y 2 meses post-tratamiento reconocen diferentes antígenos en los PES de hembras, PESH. El reconocimiento de los PESH fue mayor 2 meses post-tratamiento, en sueros de ratones con infección BIS.El reconocimiento antigénico de la IgG humana (postratamiento) fue mayor en antígenos de estadios incubados con PZQ. En los PES de hembras y machos se observó variación en la respuesta IgE luego de la incubación con PZQ, mientras que en ASH no. Se concluye que la compleja respuesta humoral humana dificulta relacionar los cambios inducidos por el tratamiento y que la incubación in vitro con la droga previa a la preparación de antígenos y el empleo de esquemas de infección murina estadio específica, permite una observación clara de la sinergia PZQ-respuesta inmune

    SECRETORY SOLUBLE ANTIGENS OF Schistosoma worms, TREATED WITH PRAZIQUANTEL The immune response and the effect of praziquantel (PZQ) on adult S. mansoni are synergistic. We evaluated the IgG and IgE in serum of mice infected with cercariae from infected snails unimiracidial in B. glabrata (UNI infection) and infected patients, soluble egg antigens and excretory-secretory products of male and female worms incubated or not with PZQ in vitro, by Western blot. The IgE in the serum of mice infected with UNI and 2 months post-treatment recognize different antigens in the PES of females, PESH. The recognition of two months PESH was higher post-treatment, sera from mice infected with BIS. The antigen recognition of human IgG (post-treatment) was higher in stage antigens incubated with PZQ. In the PES of females and males were observed variation in the IgE response after incubation with PZQ, whereas ASH no. We conclude that the complex human humoral response difficult to relate changes induced by treatment and incubation in vitro with the drug prior to the preparation of antigens and the use of murine infection patterns specific stage, allows a clear observation of synergy PZQ -immune response.
