Antimicrobial resistance represents a growing public health concern that has driven the search for therapeutic alternatives derived from medicinal plants with proven biological activity. Therefore, this study evaluated the antimicrobial potential and hematological safety of three plant extracts using the Kirby–Bauer disk diffusion method to determine antimicrobial activity against Escherichia coli (ATCC K12) and Staphylococcus aureus (ATCC Seattle 1945), with ciprofloxacin used as a positive control. Hemocompatibility was assessed through a hemolysis assay in human O+ erythrocytes, with spectrophotometric readings at 540 nm. The extracts demonstrated moderate antimicrobial activity, with inhibition halos ranging from 9–14 mm against Escherichia coli and 7–14.33 mm against Staphylococcus aureus, representing relative inhibition percentages between 20–39% compared to the positive control. Peumus boldus exhibited the greatest effect against Staphylococcus aureus, (14.33 mm). In the hemolytic evaluation, Peumus boldus showed a non-hemolytic profile (<2%), whereas Lavandula angustifolia reached 51.8% hemolysis at 3000 ppm, decreasing to slightly hemolytic values at lower concentrations. This study reveals that the evaluated extracts possess moderate antimicrobial activity and variable safety profiles, highlighting Peumus boldus as the most promising candidate.
