Evaluating Culture Media to Enhance Tissue Culture Acclimatization of Kappaphycus alvarezii

    • The tissue culture of Kappaphycus alvarezii offers a solution to limited seed availability, with the acclimatization of parental stock, supported by nutrient supplementation through culture media such as Provasoli’s Enriched Seaweed (PES) and Conway, being a critical stage for survival under controlled conditions. This study evaluated the effects of culture media type and dose on the morphology, growth, and survival of K. alvarezii parental stock during acclimatization and identified the most effective treatment. Six treatments were tested: A1 (PES, 1 mL L−1), A2 (PES, 3 mL L−1), A3 (PES, 5 mL L−1), B1 (Conway, 1 mL L−1), B2 (Conway, 3 mL L−1), and B3 (Conway, 5 mL L−1), arranged in a two-factor factorial design within a Completely Randomized Design (CRD) with three replications. Data collected included hue value, texture, bud addition, absolute growth rate (AGR), specific growth rate (SGR), and survival rate. Results showed that culture media and dose significantly influenced hue, bud addition, absolute growth, and specific growth rate, while texture and survival were statistically similar across treatments. PES at 3 mL L−1 (A2) yielded the highest bud addition (17.67 ± 1.15), whereas Conway at 3 mL L−1 (B2) produced the best numerical growth performance (AGR 1.27 ± 0.35 g; SGR 0.32 ± 0.09% day⁻¹) with stable survival (9.96 ± 0.24 g). These findings highlight the importance of selecting appropriate media formulations, with PES favoring bud induction and Conway supporting growth performance, to optimize acclimatization efficiency in seaweed tissue culture and contribute to sustainable aquaculture seed production
