    • Type I allergy is an immunological disorder which affects more than 25% of the population in the industrialized world. Allergic patients are characterized by a strong IgE-mediated Th2 response when challenged with otherwise innocuous antigens known as allergens. Treatment strategies for allergic diseases include medication aimed towards symptomatic relief and allergen-specific immunotherapy (SIT). SIT has been used since the early 1900s and involves the regular subcutaneous injection of increasing doses of allergen extract eventually leading to allergen tolerance. SIT is the only treatment approach that modifies the immune response towards allergens and is capable of long term reduction of symptom and medication scores, achieving an overall improvement in the patient's quality of life as well as decreasing the development of sensitization to new allergens. The mechanisms by which SIT exerts its beneficial effects remain incompletely defined. However, multiple studies show that SIT modifies the responses of APCs, T cells and B cells generating a Th1-Treg response that implies overall increases in IFN-¿, IL-10 and TGF-ß production. Clinical efficacy of SIT is dose dependant, a fact that involves potential side effects such as anaphylactic shock.
