Título: Efecto radioprotector del licopeno, curcumina, hidroxitirosol y melatonina sobre glándula parótida de ratas

Resumen: INTRODUCCIÓN La radioterapia constituye hoy en día un pilar importante en el tratamiento oncológico. El efecto de la irradiación puede provocar efectos no deseados sobre tejidos sanos. La radiación ionizante en las glándulas salivales produce efectos inflamatorios y degenerativos en el parénquima de las glándulas salivales, especialmente en las células acinares serosas, provocando una disminución de la tasa de secreción salival y consecuentemente problemas como infecciones orales, caries dentales, y dificultades para comer. Es un reto para la ciencia paliar estos efectos mediante sustancias radioprotectoras. OBJETIVOS El objetivo general de esta tesis es la evaluación del efecto radioprotector del licopeno, curcumina, hidroxitirosol y melatonina en las glándulas salivales parotideas de ratas Sprague Dawley a dosis única de 20Gy de radiación. Se han analizado variables histopatológicas en las glándulas parótidas como la necrosis celular, pérdida de estructura acinar, daño de conductos y vacuolización y también valores histomorfométricos como el área y perímetro de los acinos en cada uno de los grupos de estudio. MATERIAL Y MÉTODOS Se han utilizado 58 parotidas de 30 ratas Sprague Dawley (se perdieron 2 muestras del grupo control durante el procesado) divididas aleatoriamente en 6 grupos de estudio: control (n=8), licopeno (n=10), curcumina (n=10), hidroxitirosol (n=10), melatonina (n=10) y DMSO (n=10). Las diferentes sustancias fueron administradas vía peritoneal 24 horas antes de la irradiación (20Gy) y las ratas fueron sacrificadas 24 horas después de la irradiación. Las muestras fueron fijadas con Hematoxilina-Eosina y observadas bajo microscopio óptico a 40x. RESULTADOS En cuanto a las variables histopatológicas: la necrosis celular (p=0,008) y la pérdida de estructura acinar (p=0,020), es significativamente menor en los grupos de las ratas radioprotegidas, no habiendo diferencias entre los diferentes radioprotectores. En el daño de conductos (p=0,001), es significativamente menor en los grupos de las ratas radioprotegidas siendo la melatonina la que obtuvo resultados significativamente mejores con respecto a la curcumina y el licopeno. En cuanto a la vacuolización (p=0,047), el grupo del DMSO presentó un menor grado que el resto de grupos a estudio, no habiendo diferencias entre ellos. En cuanto a variables histomorfométricas, área y perímetro de los acinos, podemos decir que la melatonina, la curcumina y el hidroxitirosol tienen un mayor área y perímetro de forma significativa (p<0,001) con respecto al grupo control, licopeno y DMSO por lo que se puede deducir que a mayor área y perímetro, menor afectación del acino. CONCLUSIONES En nuestro estudio hemos observado que el grupo de ratas control (no radioprotegidas) ofrecían de forma significativa peores resultados en todas las variables a estudio con respecto a al menos uno de los radioprotectores estudiados. INTRODUCTION Radiotherapy is an important pillar in cancer treatment nowadays. The effect of irradiation may cause unwanted effects on healthy tissues. Ionizing radiation in the salivary glands results in inflammatory and degenerative effects in the parenchyma of the salivary glands, particularly in serous acinar cells, producing a decrease in the rate of salivary secretion and thus, further issues such as oral infections, dental cavities, and difficulty eating. It is a challenge for the science to relieve these effects by means of radio protective substances. TARGET The general aim of this thesis is the evaluation of the radio protective effect that lycopene, curcumin, hydroxytyrosol and melatonin produce in the parotid salivary glands of Sprague Dawley rats to the only dose of 20Gy of radiation. There were analysed histopathological variables in the parotid glands as the cellular necrosis, loss of acinar structure, damage of ducts and vacuolation as well as histomorphometric values as the area and perimeter of the acinus on each of the study groups. MATERIALS AND METHODOLOGY There were used 58 parotid glands from 30 Sprague Dawley rats (2 samples of the control group got lost during the process), which were randomly sorted in 6 groups of study: control (n=8), lycopene (n=10), curcumin (n=10), hydroxytyrosol (n=10), melatonin (n=10) and DMSO (n=10). The different substances were administered through peritoneal way 24 hours before the irradiation (20Gy) and the rats were sacrificed 24 hours after the irradiation. The samples were tinted with Haematoxylin-Eosin and observed under optical microscope to 40x. RESULTS In terms of the histopathological variables: the cellular necrosis (p=0,008) and the acinar structure´s loss (p=0,020), both are significantly less in those groups where rats received radiological protection with no differences between the substances used. With regards to ducts´ damage (p=0,001), this is considerably lower in the groups with treated rats resulting the melatonin the substance getting notably better results rather than curcumin and lycopene. As for the vacuolation (p=0,047), the DMSO group showed up with a lower level than the remaining study groups, which, in fact, did not unveil differences between them. Regarding the histomorphometric values, area and acinus perimeter, it can be stated that melatonin, curcumin and and hydroxytyrosol have a significant larger area and perimeter (p <0,001) compared to the control, lycopene and DMSO groups, therefore it can be concluded that the larger area and perimeter are, the lesser the affectation to the acinus is. OUTCOME After the research, it was noticed that the “control group” of rats (with no protection) were showing significant worse results on every variable in the survey with regards to -at least- one of the proposed substances.
